Effect of Combined Treatment with Catechin and Quercetin on Hepatic Glucose Metabolism in Diabetic Rats

Abstract
This study investigated the effects of catechin and quercetin on the hepatic glucose metabolism of diabetic rats. The experimental group was divided into five groups, the normal control (NC) group, diabetes mellitus (DM) group, diabetes mellitus+catechin (DM£«C) group, diabetes mellitus+quercetin (DM£«Q) group, and diabetes mellitus+ catechin+quercetin (DM£«C£«Q) group. Diabetes mellitus was induced by a high-fat diet and low dose streptozotocin administration. Catechin (150 mg/kg BW) and quercetin (150 mg/kg BW) were orally administered to the diabetic rats daily for three weeks. While the fasting blood glucose level decreased in the DM£«C group, DM£«Q group, and the DM£«C£«Q group compared to the DM group, the fasting blood glucose level in the DM£«C£«Q group was the lowest. Hepatic glycolytic key enzymes, glucokinase, phosphofructokinase, and pyruvate kinase activity decreased in the DM group compared to the NC group. The activity of the hepatic glycolytic key enzymes increased in the DM£«C, DM£«Q, and DM£«C£«Q groups compared to the DM group, and the enzyme activity was the highest in the DM+C+Q group. The activity of hepatic glucogenic key enzymes, glucose 6-phosphatase, fructose 1,6-bisphosphatase, and phosphoenolpyruvate carboxykinase, increased in the DM group compared to the NC group. The activity of key enzymes for hepatic gluconeogenesis decreased in the DM£«C, DM£«Q, and DM£«C£«Q groups compared to the DM group, and the enzyme activity was the lowest in the DM£«C£«Q group. The results of the above experiment show that, the combined catechin and quercetin treatment had a greater effects in improving the activity of key enzymes for glucose metabolism in diabetic livers than when administered individually.
